ABSTRACT

Objective:To investigate the regulatory role of defferentially expressed LOC107987438 in the pathogenesis of depressive disorder and provide a theoretical basis for its clinical application in depressive disorder.Methods:Differential expression of LOC107987438 was verified by quantitative real-time polymerase chain reaction(qRT-PCR)in peripheral blood monocular cells(PBMCs)of 60 patients with depressive disorder and 60 health controls. In addition, its diagnostic value was assessed by receiver operating characteristic(ROC)curves. Based on the ceRNA mechanism of lncRNA, the miRDB database was applied to predict the target miRNAs of LOC107987438, and the miRNAs with target score ≥ 80 among them were screened out.The screened miRNAs were then used to predict their potential target mRNAs through four databases which were TargetScan 8.0, miRTarBase, mirDIP and miRPathDB. Moreover, the predicted target mRNAs were annotated for gene ontology(GO)function annotation and tokoyo encyclopedia of genes and genomes(KEGG) pathway enrichment analysis via ClusterProfiler 4.0.5 package of R 4.1.1. Finally, a protein-protein interaction network was constructed using the STRING 11.5 platform to retrieve the interacting genes.Results:The qRT-PCR results showed that normalized expression of LOC107987438 in PBMCs of patients with depressive disorder was higher than that in health controls(depressive disorder: 2.084±1.357, health controls: 1.000±0.660, P<0.001). The ROC curve results showed that the area under curves(AUC)of LOC107987438 was 0.759(95% CI: 0.675-0.842, P<0.05), indicating its high potential diagnostic value. Bioinformatics analysis showed that hsa-miR-4670-3p, hsa-miR-619-3p, hsa-miR-6721-5p and hsa-miR-297 were the miRNAs with high bindings to LOC107987438. The results of KEGG signaling pathway enrichment revealed that hypoxia-inducible factor 1(HIF-1)signaling pathway, phosphatidylinositol 3-kinase-AKT(PI3K-Akt) signaling pathway and erythroblastic oncogene B(ErbB) signaling pathway were closely associated with depressive disorder. Among the top ten key genes screened by the protein-protein interaction network, kirsten rats arcomaviral oncogene homolog(KRAS), androgen receptor(AR), cyclic-AMP response binding protein1(CREB1), insulin-like growth factor 1(IGF1), cyclin-dependent kinase inhibitor 1B(CDKN1B) and calcium/calmodulin-dependent protein kinase type Ⅱ alpha(CAMK2A)were strongly associated with depressive disorder. Conclusion:The establishment of ceRNA regulatory network of LOC107987438 provides a theoretical basis for exploring the pathophysiology of depressive disorders.

ABSTRACT

Objective:To analyze the application effects of anchored instruction combined with problem-based learning (PBL) in the clinical teaching of department of gynecology.Methods:Taking the implementation time (June 2020) of anchored instruction combined with PBL in the hospital, 86 clinical interns who entered the department of gynecology before the implementation (from April 2019 to May 2020) were included in control group, and this group adopted traditional teaching method; 82 interns enrolled after the implementation (between June 2020 and December 2020) were selected as observation group and were given new teaching method. The theoretical assessment results, practical operation results, independent learning ability and teaching satisfaction were compared between the two groups after 1 month of training. SPSS 22.0 was used for t test and rank sum test. Results:After 1 month of training, the scores of theoretical assessment results [(81.20±10.64) vs. (73.96±8.25)] and mini-clinical evaluation exercise (Mini-CEX) scale of observation group were significantly higher than those of control group, and the independent learning ability and teaching satisfaction evaluation were significantly better than those of control group ( P<0.05). Conclusion:Anchored instruction combined with PBL can effectively improve the independent learning ability and learning satisfaction of interns in department of obstetrics and gynecology, with good application effects.

ABSTRACT

In recent years, with the development of sequencing technology and research on molecular changes in different races of prostate cancer, it has been found that the pathogenesis of prostate cancer showed obvious ethnic differences. This article reviewed the research progress of ETS fusion genes, FOXA1, SPOP, IDH1 and other driver genes in prostate cancer. And the research of molecular typing of prostate cancer showed different patterns of molecular changes in Chinese and western populations. The molecular changes of prostate cancer in western populations were dominated by ETS fusion gene, while those in Chinese populations were dominated by gene mutations, mainly FOXA1 and SPOP mutations. Moreover, the dominant fusion gene in Chinese prostate cancer was not ETS fusion gene, but SCHLAP1-UBE2E3.

ABSTRACT

Objective To observe the clinical effects of the adjustive tractor for cervical dislocation.Methods Forty-seven patients were included between September 2007 and November 2011.There were 36 males and 11 females with age ranged from 7 to 62 (mean,35 years).The mean interval from injury to admission was 8.6 h (range,0.5-72 h).There were atlanto-occipital dislocation in 2 cases,C1.2 in 2 cases,C2.3 dislocation in 5 cases,C3.4 dislocation in 2,C4.5 in 9,C5.6 in 13,C6.7 in 14.Thirty cases were complicated by fracture.No facet locking occurred in sixteen cases.Facet locking was found in ten cases and bilateral facet locking was in 21 cases.After reduction,brace or internal fixation followed.According to American Spinal Injury Association (ASIA) spinal function impairment scale standard,there were 4 cases in level A,10 cases in level B,18 in C,10 in D,and 5 in E.According to Japanese Orthopaedic Association (JOA) spinal function rating standard,the mean JOA score was 9 (range,2-14).Results All 47 cases were reduced successfully without neuronal function aggravation.Traction power ranged from 7 to 60 kg (mean,25.6 kg),the mean time of traction was 8 min (range,3-10 min).The mean follow-up was 38 (range,6-48) months.All the patients achieved normal alignment and intervertebral height.The intervertebral body fusion was observed in all of cases,the mean fusion time was 3.3 months (range,3-6 months).One patient who experienced nonunion of vertebral arch fracture didn't receive further treatment because of absence of symptoms.At last follow-up,there were 3 cases in level A,1 in level B,4 in C,8 in D,and 31 in E according to ASIA scale.The mean JOA score was 12 (range,2-17).Conclusion The adjustive tractor is simple and safe for prompt reduction.

ABSTRACT

BACKGROUND: Bone marrow mesenchymal stem cells are prospective used in cartilage tissue engineering due to easy acquire and plentiful amplification in vitro in a short term.OBJECTIVE: To summarize the application of bone marrow mesenchymal stem cells in repairing cartilage defect.RETRIEVAL STRATEGY: A computer-based online search was conducted in Pubmed for English language publications containing the key words of "marrow; mesenchymal stem cells; cartilage defect" from October 1982 to December 2006, Relevant data were also searched in China Scientific and Technological Achievement Database for Chinese language publications containing the key words of "marrow; mesenchymai stem cells; cartilage defect" from October 1982 to December 2006. There were 126Exclusion criteria: duplicated articles.LITERATURE EVALUATION: Literatures including reviews and experimental studies were mainly derived from Pubmed database and China Scientific and Technological Achievement Database.DATA SYNTHESIS: It has been proved that bone marrow mesenchymal stem cells could differentiate into cartilage in vivo.However, differentiation of cartilage phenotype in vitro was restricted by multiple factors, and the controlling mechanism is still unclear up to now. Animal experiments demonstrated that bone marrow mesenchymal stem cells could repair bone and cartilage defect. Although studies on bone marrow mesenchymal stem cells and cartilage tissue engineering have developed to a certain degree, clinical applications and evaluations, including cell marks of bone marrow mesenchymal stem cells in different differentiated stages, controls of proliferation and differentiation of bone marrow mesenchymal stem cells, and gene transfection technique, still need a further study.CONCLUSION: Animal experiments indicate that bone marrow mesenchymal stem cells can significantly repair bone and cartilage defect. Although studies on bone marrow mesenchymal stem cells and cartilage tissue engineering have developed to a certain degree, problems of bone marrow mesenchymal stem cells in basic and clinical researches still need to be solved further.

ABSTRACT

Objective To study the flow fields of electrolysis oxygen generation with micro-flow-channels of small square cylinders and the key structure factors impacting on the uniform distribution of the water flow in the micro-flow-channels.Methods The low Renault k-? model was used to represent the characteristics of the flow field in the micro-flow-channels.And the simulation results of the flow fields by CFD were calibrated by comparing with that of the particle image velocimetry(PIV) experiments,which showed that the low Renault k-? model was suitable to describe the flow field of the micro-flow-channels.Results The flow fields of variable flow were obtained.Conclusion The flow fields of variable flow rates distribute non-uniformly.The velocity magnitude is 10-3m/s,and the inlet structure of the electrolysis oxygen generation is a key factor impacting on the uniform distribution of the water flow.
